Tissue Culture Studies in Tomato (lycopersicon Esculentum) Var. Moneymaker

چکیده
A protocol was developed for callus induction and regeneration in tomato (Lycopersicon esculentum) var. Moneymaker. Hypocotyl and leaf disc explants of tomato were used as a starting material for callus induction. Explants were cultured on MS medium having different concentrations of hormones. Maximum callogenesis from hypocotyls was obtained on MS medium supplemented with IAA (2 mg/l), NAA (2 mg/l), BAP (5 mg/l) and Kin (4 mg/l). It was 65.2% for hypocotyls. For leaf discs maximum callogenesis was achieved on MS medium supplemented with IAA (2 mg/l), NAA (2 mg/l), BAP (2 mg/l) and Kin (4 mg/l). It was 81.3% for leaf discs. Calli were cultured on MS medium having concentrations of Zeatin (1 mg/l) and IAA (1 mg/l) for regeneration. They showed maximum regeneration of 69.2% from hypocotyls. Minimum regeneration of 2.8% was obtained on MS medium supplemented with IAA (2 mg/l), NAA (2 mg/l), BAP (2 mg/l) and Kin (4 mg/l). Tomato shoots were shifted to 1⁄2 MS medium containing IBA (0.1 mg/l) and BAP (0.0025 mg/l) for rooting and all responded positively to rooting.

Lipoxygenase pathway and antioxidant system in salt stressed tomato seedlings (Lycopersicon esculentum Mill.)
Soil or growing media salinity is an increasing problem in agriculture throughout the world. Therefore, there is a real interest in a better understanding of the mechanisms set by the plants to respond to this stress. The present study concerns tomato, which is one of the most important cultivated vegetables. Tomato seeds (Lycopersicon esculentum var. 'Moneymaker') were disinfected and a salt s...
متن کاملPrimary dormancy in tomato (Lycopersicon esculentum cv. Moneymaker): studies with the sitiens mutant
Intact wild-type tomato (Lycopersicon esculentum cv. Moneymaker) seeds do not complete germination to the same percentage or at the same speed as intact ABA-deficient sitiens (sit") mutant seeds when seeds of both genotypes are imbibed on polyethylene glycol (PEG) solutions of -0 .3 to —1.5 MPa osmotic potential. However, if the thicker testas of wild-type seeds are removed (stripped) from the ...
متن کاملThe effect of seed priming and culture medium type on germination characteristics and quality of tomato seedlings (Lycopersicon esculentum)
Extended Abstract Introduction: Today, seed priming is widely used to improve seed germination in a wide range of plant species. As a result of seed pre-treatment, several molecular and biochemical changes occur, including increased macromolecule synthesis, enzyme activity and formation of different metabolites. Enzymatic and metabolic activities, synthesis of proteins in quantitative and qual...

this study sought to investigate the effects of two tomato varieties [Lycopersicon esculentum Mill. var. esculentum (Esc) and Lycopersicon esculentum Mill. var. cerasiforme (cEr)] on acetylcholinesterase (AchE) and butyrylcholinesterase (bchE) activities in vitro. Phenolics content, carotenoids characterisation, inhibition of Fe2+ and quinolinic acid-induced malondialdehyde (MDA) production in ...
